Ubiquitin (Ub) is a 76-amino-acid protein that mediates post-translational modification of target proteins through the E1–E2–E3 ubiquitination cascade. Ub can be attached to substrates as monoubiquitin or polyubiquitin chains, regulating diverse cellular processes including proteasomal degradation, protein trafficking, and signal transduction. Polyubiquitinated proteins are often targeted to the 26S proteasome for degradation. This recombinant ubiquitin protein contains an N-terminal 6×His tag and a thrombin cleavage site for convenient purification and downstream applications. Recommended concentration for in vitro ubiquitination reactions is 50–100 μM.
